The caloTrace® is Symcel’s most flexible biocalorimetry platform, designed to turn real-time metabolic monitoring into a powerful solution for microbial QC testing, research, and product development across a wide range of applications. It combines ease of use with flexible sample volumes and exceptional measurement sensitivity. Samples can be incubated at temperatures between 20 and 40 °C across 30 independently loaded test positions with continuous monitoring and automatic pass/fail criteria.
NEXT-GENERATION BIOCALORIMETRY SYSTEMS
Expanding on the proven calScreener® platform, the caloTrace® detection system utilizes highly sensitive isothermal microcalorimetry technology, detecting the heat released by living cells and microorganisms with exceptional sensitivity.
There is no need for reagents, enrichment, filtration, lysis, or other disruptive sample preparation steps. Even complex sample types, including creams, powders, food samples, liquids, and solid materials, can be tested directly in sample volumes of up to 10 mL.
Operating at controlled temperatures from 20 to 40°C, the system accommodates a broad range of applications, from microbial and cellular metabolism studies to formulation, stability, and reaction monitoring. RAPID, AUTOMATED AND SIMPLIFIED MICROBIAL MEASUREMENTS
The caloTrace sets a new standard for microbial testing, introducing a number of new benefits. High-sensitivity detection of bacteria, yeast, and mold combined with rapid and continuous monitoring enables operators to take immediate action as soon as microbial presence is detected.
Samples, including complex and difficult matrices, can be analyzed directly in test vials with standard media without filtration, reagents, or cumbersome and error-prone handling steps. The system enables testing of up to 30 samples in parallel, with each position loaded independently for maximum flexibility and throughput. Clear automated pass/fail results remove subjective interpretation errors, while detection of only viable microorganisms minimizes the risk of false results.

METABOLIC ANALYSIS OF COMPLEX BIOLOGICAL SYSTEMS
Based on isothermal microcalorimetry, the system expands the flexibility of biocalorimetry and metabolic monitoring for testing and research of organic materials. Sample volumes of up to 10 mL can be analyzed across a wide range of applications, including mammalian cells, tissue biopsies, yeasts, fungi, algae, plants, grains, and soil samples.
Conventional techniques frequently require extensive sample preparation, optical clarity, or sample disruption, which slows workflows and increases the risk of non-representative results. Since heat production is an intrinsic property of life, biocalorimetry enables direct, in situ, continuous, and non-destructive biological detection without altering the sample.
STREAMLINED WORKFLOW
The caloTrace combines sensitive measurement with practical sample handling. Using autoclavable crimp-cap glass vials, the system accommodates both liquid and solid samples prepared by weighing, pipetting, or direct injection. Add 10 mL sample or sample-media mix to a 13 mL vial – if less than 10 mL, fill up with media – cap vial. Once sealed, samples are monitored non-destructively in real time, preserving material integrity and allowing retrieval for follow-up characterization or further analytical testing.
The simplicity of the workflow not only makes testing easier for operators, but also improves reliability and reproducibility. The simpler an analytical method is, the lower the risk of operator errors and the more reliable the results become.
Independent Channels, reliable Data
Featuring 30 independent isothermal calorimetric channels equipped with high-performance heat flow detectors, the caloTrace is designed for stable and reliable operation. Each detection unit contains a built-in reference and no integrated moving parts, ensuring long-term reliability while minimizing the risk of downtime.
The sensors in each channel are extremely sensitive, capable of capturing even very small amounts of metabolic heat released by microorganisms, enabling single-CFU limits of detection. Unlike systems that rely on multiple secondary optical measurements, the caloTrace provides a more direct and rapid microbial detection method unaffected by sample viscosity or variability in optical clarity.
CALVIAL - PRECISION SAMPLING
Measurements are performed using autoclavable caloTrace 13 mL calVials™ with a total sample volume of up to 10 mL. Samples can either be added through the septum or filled directly into the vial prior to sealing with a crimping tool using autoclavable crimp-cap glass vials.

The calView® software interface streamlines the entire workflow—from sample entry to result reporting. It allows real-time monitoring of all measurements and offers flexible data visualization, from full experiment overviews and data exports to individual sample curves. It transforms complex thermal data into intuitive, interpretable results for confident decision-making.
For QC microbial testing applications it can se set up to provide automatic positive/negative results and analysis reports. The system supports 21 CFR Part 11 data integrity requirements with audit logs, user roles, and secure electronic documentation.

The caloTrace detection technology is based on the principle of biocalorimetry using isothermal microcalorimetry (IMC). IMC continuously detects the metabolic heat released by organisms in the test vials at sub-microwatt (µW) levels, under controlled set incubation temperatures.
Unlike methods relying on visible growth or optical signals, caloTrace directly measures metabolic heat, enabling earlier and more sensitive detection without waiting for indirect markers. The caloTrace method is label-free and can test almost any sample directly in the vial without filtration or extensive sample preparation. Unlike molecular methods, caloTrace detects only viable microorganisms, minimizin the risk of false results. Continuous real-time monitoring also enables immediate action when microbial growth above a defined threshold is detected.
The caloTrace is based on IMC, enabling exceptional flexibility in sample types, including complex, solid, or viscous formulations. Applications range from consumer goods, cell-based samples, and organic materials to challenging industrial and food-related samples where traditional methods often struggle. If it fits in the cup, you can measure it.
The caloTrace operates between 20 and 40 °C and supports up to 10 mL sample volumes with independently loaded sample channels. The calScreener and calScreener+ measure up to 32 batch-loaded samples of 600 µL each. The calScreener operates between 30 and 40 °C, while calScreener+ operates between 20 and 40 °C. The calScreener+ Sterility is a packaged solution configured for sterility testing aligned with dual-temperature compendial testing requirements.
The caloTrace is designed for simple and intuitive operation with minimal hands-on time. Samples are prepared directly in standard glass vials, loaded individually into the system, and monitored automatically in real time through the calView software. The streamlined workflow minimizes manual handling and complex sample preparation.
The caloTrace system is designed to require minimal maintenance. No routine user maintenance is required beyond standard system cleaning. Annual preventive maintenance performed by a certified service technician includes instrument calibration, system verification, and, when applicable, software updates.
